In4mer Library Generator

Generate combinatorial Cas12a guide RNA libraries for genetic interaction screening. Each construct contains a 4-mer array of crRNAs for enAsCas12a.

Library Design

  • Single knockouts: 2 targeting guides + 2 intergenic padding guides, 3 clones per gene
  • Pairwise knockouts: 2 guides per gene (4 total), 3 clones per pair
  • Reference controls: Essential + nonessential genes (customizable)
Library size: 1.5n(n+1) + 3(E+N) clones
where n = target genes, E = essential genes, N = nonessential genes

2. Target Genes

Drop file to load genes
One gene per line, or separated by commas/tabs. Accepts .txt, .csv, .tsv files

Include Known Synthetic Lethal Pairs

Select paralog pairs to add as positive control synthetic lethals.
